Modulation of mRNA stability as a powerful tool plays an important role in controlling gene expression during the physiological transitions and regulatory processes. Novel strategy to characterize mRNA stability

So far, mRNA stability and turnover have been measured by various approaches, such as using actinomycin D, metabolic labeling. The use of actinomycin D is a classical method, which is able to determine the decay in RNA abundance over time following inhibition of transcription. Metabolic labeling is based on metabolic labeling of RNA transcripts with modified nucleotides, allowing less disruption to cellular physiology. Each of these methods has its own advantages. However, many of them have limited sensitivity for low-abundance transcripts and the measurement results vary greatly across assays. Recently, the analysis of high-throughput RNA-seq data is emerging as a promising method for estimation mRNA stability. The novel strategy is based on intronic reads, producing a relative measure of RNA half-life under the assumption of equilibrium conditions between RNA production and decay. Compared with the existing methods based on metabolic labeling or transcriptional inhibition, the approach is efficient, relatively non-disruptive and so on.
