Liposomes remain one of the most versatile nanocarriers for delivering genetic therapies, small molecules, and biologics. Our service portfolio covers the entire development cascade: from lipid selection and encapsulation method optimization to comprehensive physicochemical characterization.
Working with both early discovery and late-stage preclinical programs, we tailor size, charge, lamellarity, and surface ligands to match your intended route of administration and target tissue. Each project is executed under defined quality controls and documented in a detailed Certificate of Analysis.
Whether you need a proof-of-concept batch for in vitro screening or a scaled-up lot for animal studies, our team designs formulations that balance stability, encapsulation efficiency, and release kinetics. All characterization data — particle size, polydispersity, zeta potential, encapsulation efficiency, and stability profiles — are included in the final report.
Key Features
Cargo-Specific Formulation Design
We develop liposomal formulations optimized for the physicochemical properties of your cargo: small molecules, peptides, nucleic acids (siRNA, mRNA, plasmid DNA), or protein complexes (Cas9 RNP). Lipid composition, cholesterol ratio, and surface charge are systematically varied to maximize loading and retention.
Encapsulation Method Optimization
We employ thin-film hydration, ethanol injection, and reverse-phase evaporation methods, then select and refine the technique that best suits your cargo stability and target particle size. Process parameters such as hydration temperature, extrusion cycles, and lyoprotectant addition are scoped to meet project-specific encapsulation efficiency targets.
Surface Functionalization and Targeting
Ligands such as PEG, antibodies, peptides, folate, or aptamers can be conjugated to the liposome surface to prolong circulation or enable active targeting. We provide PEGylated, cationic, anionic, and stimuli-responsive formulations (pH-sensitive, thermosensitive) as specified in your project scope.
Comprehensive Physicochemical Characterization
Characterization is performed using DLS, NTA, Zetasizer, HPLC, and fluorescence/UV-based assays to measure mean diameter, PDI, zeta potential, encapsulation efficiency, drug loading, and in vitro release profiles. Stability under storage and physiological conditions is assessed over a scoped time frame.
Payloads Typically Encapsulated
Our encapsulation platform accommodates a wide range of genetic and non-genetic payloads. Below are the most commonly requested modalities; we welcome inquiries for atypical cargos.
Plasmid DNA: Encapsulation of plasmid DNA encoding gene-editing enzymes or therapeutic genes. Lipid formulation is optimized for high entrapment and protection from nuclease degradation.
mRNA: Liposomal encapsulation of in-vitro-transcribed mRNA for transient protein expression. Formulations are designed to prevent RNase-mediated degradation and facilitate endosomal escape.
siRNA / miRNA: Cationic and ionizable lipid formulations are used to complex small RNAs with high loading efficiency, enabling potent gene silencing in vitro and in vivo.
CRISPR-Cas9 RNP: Ribonucleoprotein complexes (Cas9 + sgRNA) are encapsulated in lipid nanoparticles for direct delivery, reducing off-target effects associated with plasmid-based systems.
Small Molecule Therapeutics: Hydrophilic and lipophilic drugs are loaded into the aqueous core or lipid bilayer to improve solubility, control release, and enhance bioavailability.
Workflow
Each project follows a structured, iterative process that balances scientific rigor with timely delivery. The number of optimization rounds and analytical endpoints is defined in the initial project plan.
1. Cargo and Target Profiling: We review your cargo's physicochemical properties (molecular weight, charge, stability), intended route of administration, and desired release kinetics. Based on this, we propose an initial lipid composition and encapsulation method.
2. Formulation Screening and Optimization: A panel of 5–10 formulations is prepared and screened for particle size, PDI, zeta potential, and encapsulation efficiency. Lead formulations proceed to iterative refinement (e.g., adjusting lipid ratios, extrusion conditions, or lyoprotectant).
3. Scale-Up and Reproducibility: Once the optimized formulation is confirmed at bench scale, we produce a larger batch (typically 10–200 mL, scoped per project) and verify batch-to-batch reproducibility across three independent preparations.
4. In Vitro Release and Stability Testing: Release profiles are measured under simulated physiological conditions (PBS, serum-containing media) using dialysis or ultrafiltration. Stability over time is assessed at 4°C, room temperature, and freeze-thaw cycles as specified.
5. Final Characterization and Deliverables: A complete Certificate of Analysis is generated, reporting size, PDI, zeta potential, encapsulation efficiency, drug loading, release kinetics, and stability data. Residual solvent and endotoxin levels are included if required.
Integrated Analytical Platform
All characterization is performed using calibrated, qualified instruments. Data are reviewed by a senior scientist and provided in a format suitable for internal reports or regulatory submissions.
Quality Assurance
Every liposome batch is produced under documented standard operating procedures. All characterization instruments are calibrated and maintained per our quality system. Raw data, instrument logs, and calculation sheets are retained and available for audit.
We do not guarantee a specific encapsulation efficiency or stability shelf life as these depend heavily on the cargo and intended use. Instead, we provide complete transparency: all measured values, acceptance criteria (if provided by client), and deviations are reported. For studies requiring GLP-like documentation, additional quality measures can be implemented under a separate project scope.

Target Cell Types for Liposomal Delivery
Liposome formulations are routinely tested in relevant cell models during the development phase. The table below lists common target cell categories; custom cell lines can be accommodated upon request.
Adherent Cell Lines: HEK293, HeLa, A549, HepG2, and other standard lines used for initial transfection efficiency and toxicity screening.
Suspension Cells: Jurkat, K562, THP-1 – liposome formulations with optimal surface charge and size are developed for efficient uptake in non-adherent cultures.
Primary Cells and Stem Cells: Human primary fibroblasts, mesenchymal stem cells (hMSCs), and iPSCs – low-toxicity formulations designed to preserve cell viability and differentiation potential.
Hard-to-Transfect Cells: Neurons, macrophages, dendritic cells – targeted liposomes (ligand-conjugated) or fusogenic formulations are used to enhance delivery.
