Custom siRNA Design and Synthesis

Overview

Accelerate your RNAi research with custom-designed siRNAs optimized for potency, specificity, and stability.

Small interfering RNAs (siRNAs) are powerful tools for sequence-specific gene silencing, enabling researchers to dissect gene function, validate therapeutic targets, and develop RNA-based therapies. Our siRNA design and synthesis service combines advanced bioinformatic algorithms, high-quality chemical synthesis, and rigorous quality control to deliver siRNAs that meet the demands of both basic research and preclinical development.

Poorly designed siRNAs often result in inconsistent or irreproducible knockdown, wasting valuable research resources. We address these challenges through intelligent sequence design that minimizes off-target effects, improves nuclease resistance, and enhances transfection efficiency. Our platform integrates structural accessibility prediction, thermodynamic optimization, and off-target filtering to generate siRNA candidates with high silencing activity and specificity.

From individual sequences to large-scale libraries and chemically modified conjugates, our service is modular and scalable. Whether you need a single siRNA for target validation or a genome-wide library for high-content screening, we provide the technical expertise and quality assurance to support your RNAi workflow.

Key Features

Custom siRNA Design and Synthesis

Our design service uses proprietary algorithms to analyze target mRNA structure, GC content, and thermodynamic properties, ensuring high silencing efficiency and minimal off-target potential. Each sequence is synthesized on automated solid-phase platforms under stringent quality controls. Deliverables include lyophilized or solution-form siRNA, with optional annealing into duplexes.

Chemical Modifications and Conjugates

To improve stability and delivery, we offer a wide range of chemical modifications including 2′-O-methyl, 2′-fluoro, locked nucleic acids (LNA), and phosphorothioate backbones. We also provide covalent conjugates with lipids, peptides, or fluorescent dyes (FAM, Cy3, Cy5) for tracking and targeting. Modified siRNAs exhibit enhanced nuclease resistance and improved pharmacokinetic profiles.

High-Throughput Libraries and Screening

Predesigned siRNA libraries targeting specific pathways, disease gene families, or entire genomes are available for large-scale functional screens. Our screening service integrates automated liquid handling, high-content readouts, and data analytics to rapidly identify the most effective silencing candidates. This accelerates target discovery and gene validation studies.

Quality Control and Validation

Every siRNA batch undergoes rigorous QC including HPLC or PAGE purity analysis, LC-MS identity confirmation, and endotoxin testing. Annealing efficiency is verified before shipment. For custom projects, optional knockdown validation in a cell line of choice can be included. A comprehensive QC report accompanies each order, ensuring reproducibility across experiments.

Product Formats

Our siRNA service covers a range of formats to match different experimental needs. From single sequences to pooled libraries, each format is produced with consistent quality and documentation.

Workflow

Our workflow proceeds through five phases, from target analysis to final product delivery.

Quality Assurance

Each siRNA product is subjected to a set of standardized QC assays to ensure batch-to-batch consistency and reliable biological performance. Purity is assessed by reversed-phase HPLC or denaturing PAGE, with a minimum purity threshold defined per project (typically ≥90% for standard products). Identity is confirmed by LC-MS mass spectrometry, verifying the exact molecular weight of the full-length oligonucleotide. Concentration is determined by UV absorbance at 260 nm, and annealing efficiency for duplex formulations is verified by gel electrophoresis or ion-exchange chromatography.

For modified siRNAs, additional QC may include fluorescence verification (for labeled products) and endotoxin testing (LAL assay) for in vivo applications. All QC data are compiled in a Certificate of Analysis provided with the shipment. Optional third-party validation (e.g., knockdown efficiency in a target cell line) can be scoped as an add-on service.
